The burden of chronic cough often goes far beyond the cough itself for patients living with idiopathic pulmonary fibrosis (IPF) and non-IPF interstitial lung diseases (non-IPF ILDs). From having to stop working, to the physical pain that comes from coughing, to removing themselves from everyday activities like going out to dinner or to the movies, the toll cough has had on their lives is evident. Thrilled to share our new paper in the Journal of Human Immunity. 🧬 By examining exomes from the Penn Medicine Biobank, we found a surprisingly high prevalence of STING gain-of-function variants in adults. Unlike classic SAVI — characterized by severe early-onset disease and high mortality — these adult carriers showed a spectrum of penetrance, ranging from asymptomatic to overt autoimmune disease. Adult-onset SAVI is hiding in plain sight, and our findings show that common autoimmune and autoinflammatory diseases can be caused by STING gain-of-function mutations. Inhibrx Biosciences, Inc. has reported updated interim results from its Phase 1/2 study of #ozekibart plus FOLFIRI produced a 20% objective response rate and an 87% disease control rate in heavily pretreated metastatic #colorectalcancer, where historical response rates are typically 1% to 6%. In 45 evaluable patients, the median progression-free survival was 5.5 months, and 42% remained progression-free at six months. The regimen also showed a manageable safety profile, with mostly Grade 1 or 2 treatment-related adverse events. https://lnkd.in/eEAEnkc4
